Is that a diode laser? Those won’t touch clear acrylic without a surface treatment.

@JimNM is correct, you need something on the front or back that can become a heat spot for the laser. Black paper, acrylic paint, or other materials are often used. Results depend on the coating thickness and what is used.
You will soon accept experimentation is a major component of laser work.
